本研究对金樱子组织培养快速繁殖技术进行了探索。以金樱子的带芽茎段及叶片为材料,系统地研究了金樱子组织培养间接器官发生途径的各个培养阶段中基本培养基、生长调节剂和活性炭等因素的影响,筛选出了各阶段的适宜培养基,成功地获得了金樱子再生植株,建立了系统的金樱子植株再生体系。 以金樱子茎段为外植体,采用0.1%HgCl2浸泡5~10 min,然后用青霉素和链霉素20mg/L混合液浸泡30 min,能取得较为理想的灭菌效果。3月下旬到4月上旬是进行金樱子茎段培养取材的最佳时期。MS比WPM和B5更适应金樱子芽萌芽的培养。金樱子初代培养在MS+6-BA2.0mg/L +NAA0.05 mg/L上时萌发的效果最好,腋芽萌发率最高,达到90%。 对金樱子无菌苗增殖条件进行了研究,在单独使用生长素处理时无菌苗不增殖,能够生成愈伤组织或者根。6-BA、KT、ZT这3种细胞分裂素进行比较,6-BA较其他两种更有效,且浓度为1.5mg/L时增殖系数达到3.0。MS+NAA0.15mg/L+BA1.0mg/L+蔗糖35 g/L是适合金樱子继代增殖的培养基,其增殖系数达到4.15。 无根组培苗的生根阶段,MS、2/3MS、l/2MS和1/4MS四种基本培养基,都可以生根,但以1/2MS对生根影响最佳。IBA比NAA、IAA、2.4-D对金樱子无菌苗生根效果好,0.1mg/L IBA时生根率达到94.3%。培养基中添加活性炭对生根效果不显著。 .....

The study was mainly to probe an effieient way through tissues culture and rapid propagation of Rosa laevigata Mickx.The leaves and stems with axillary buds were taken as explants. The factors that affect every step including basic mediurn, hormone and active carbon were detailed investigated through the indirect way. As a result, the optimun mediums of every step were screened.A plant regeneration system of Rosa laevigata Mickx. was established in this research.The main results are below: Establishment of aseptic culture was conducted,when sterilized by conventional way,stems were sterilized by immersed 5~10 min with mercuric chloride(0.1%)first,and then immersed 30 min with penicillin and streptomycin 2 million units,the contamination rate was 13.3%.The best time for explants acquisition was in late March to early April. For germination of Rosa laevigata Mickx. , MS was propitious to WPM or B5.The germination rate with 2.0mg/L 6-BA and 0.05 mg/L NAA was suitable to germinate of Rosa laevigata Mickx., the germination rate could reach 90%. Axillary buds proliferation was studied.The effeets of different auxins were indicated that: NAA、IBA、IAA can only inducted callus or roots.At the same concentrations,6-BA is more efficient than KT and ZT.The optimum culture medium of proliferating axillary buds was MS+NAA0.15mg/L+BA1.0mg/L.The multiplication rate was 4.15. ......
