大豆[Glycine max (L.) Merr.]作为主要的油料作物,在农业生产和食品工业中占有重要地位。大豆种子是植物油和蛋白的重要来源之一,其相关研究受到人们越来越多的关注。发掘和研究大豆中调控种子形态发育的基因是大豆生长发育分子生物学研究的重要基础工作,而相关突变体的获得则提供了良好的遗传材料。本文中以诱变方式获得的大豆子叶折叠突变体为材料,筛选出与子叶发育相关的两个基因:GmGDSL1与GmDof17-1,并通过转基因技术确定其功能。
本研究采用NaN3-60Coγ诱导处理大豆“南农94-16”种子,获得子叶折叠突变体,并对其进行系统性分析。突变体M8代子叶折叠表型保持稳定,该性状出现在子叶形态发生期,且一直保持到种子成熟期。与对照材料相比,突变体除了子叶保持折叠状态外,还出现了植株矮化,成熟期延迟,侧根数目减少等性状,其它性状则没有明显差异。品质测定表明:突变体蛋白质含量明显提高,达到52%,而对照材料的蛋白质含量只有44%;突变体油脂含量仅有16.6%,而对照材料则有17.8%;氨基酸分析发现:突变体种子中氨基酸含量明显高于对照材料种子中的含量,尤其是含硫氨基酸含量明显提高。细胞学观察结构表明:突变体种子内部分细胞器降解,液泡内出现多膜体结构,其液泡也明显大于对照种子的液泡。生长素含量检测表明突变体内源生长素(IAA)含量明显降低。

Soybean [Glycine max (L.) Merr.] as a major economic crop plays an important role in agricultural production and food industry. It is one of the most important resources of plant oil and protein for human beings, seed development in soybean attracts more and more attention. It is an important foundation for molecular biology study on soybean soybean growth and development to explore and study the morphological development of soybean seeds, and the relevant mutant provides a good access to good genetic materials. In our paper, we take advantage of curled-cotyledons mutants in soybean treated by compound mutagenesis, and find two genes related to cotyledon development, GmGDSL1 and GmDof17-1 by using a variety of experimental methods.
In our research, we obtained a recessive soybean mutant named curled-cotyledons, which is derived from soybean cultivar ‘Nannong 94-16’ treated by NaN3 and 60Coγ rays. The curled cotyledons trait was stable in M8 generation plants, which appeared in the early morphogenesis, and maintained until seeds matured. Results of character survey showed that mutants exhibited dwarf plant, later maturity and decreased number of lateral root except curled cotyledons trait. Quality testing showed that protein content was significantly higher in curled-cotyledons mutants, up to about 52%. On the contrary, the oil content was decreased in mutants, only 16.6%, whereas was 17.8% in wild type.
